[Pharmacological Action]
Pharmacology
Spectinomycin belongs to the class of aminoglycoside antibiotics, and has a moderate inhibitory effect on a variety of Gram-negative bacilli, such as E. coli, Salmonella, Shigella, and Proteus. It is sensitive to streptococcus, pneumococcus, staphylococcus epidermidis and certain mycoplasmas (such as Mycoplasma gallisepticum, Mycoplasma turkeyi, Mycoplasma synovialis, etc.). Mostly insensitive to Streptococcus aureus and Staphylococcus aureus. It is usually resistant to Pseudomonas aeruginosa and Treponema pallidum. Intestinal bacteria are more resistant to spectinomycin, but they do not exhibit cross-resistance with streptomycin. Lincomycin has good antibacterial activity against anaerobic bacteria, such as Clostridium, Peptococcus, Peptostreptococcus, Clostridium tetanus, Clostridium perfringens and most actinomycetes. Lincomycin mainly acts on the 50S subunit of bacterial ribosomes and affects protein synthesis by inhibiting the elongation of peptide chains.
Pharmacokinetics
Spectinomycin only absorbs 7% after oral administration, but maintains a higher concentration in the gastrointestinal tract. The tissue concentration of the drug is lower than the serum concentration. It is not easy to enter the cerebrospinal fluid or eyes, and the rate of binding to plasma proteins is not high. Most of the drug is discharged through glomerular filtration in its original form. Lincomycin is poorly absorbed internally and has a bioavailability of 30% to 40%. Mixed feeding can reduce its absorption speed and absorption. Chickens took 50 mg of this product per kilogram of body weight (dissolved in drinking water) for 7 days. [Pharmacological interaction]
(1) If used together with Lincomycin, it can significantly increase the antibacterial activity against Mycoplasma and expand the antibacterial spectrum.
(2) If Lincomycin used together with Anticholinesterase, and it might decrease the effect of latter one.
(3) If used together with Erythromycin, there might be Antagonism.
